Bispecific IgG (BsIgG), IgG with an extra antigen-binding moiety attached, bispecific fusion proteins, BsAb fragments, and BsAb conjugates are the five main structural subgroups of BsAb. Each of these several BsAb forms handles certain aspects of each antigen's binding valency, pharmacokinetic half-life, and antigen-binding site shape. By adding extra antigen-binding units to either the amino or carboxy termini of either the light or heavy chains, monospecific IgG can be made to be bispecific. There are a number of additional antigen-binding units available, including paired antibody variable domains, designed protein scaffolds, and single domain antibodies (unpaired VL or VH) (e.g., Fv or scFv). One of the appended IgGs, KIH IgG-scFab, is offered for customization by Creative Biolabs.
